      During my Ph.D training I gained hands on experience in expression and purification of membrane proteins from E.coli media, involving ultracentrifugation, membrane solubilization, Ni-NTA histidine tag purification, FPLC and SDS-PAGE. I have also used techniques such as Circular Dichroism and Thermaflour Assay for protein secondary structure determination. My main focus however was using NMR to establish the global fold of the proteins in solution as well as identify protein-ligand interaction sites. I used solid-state NMR to characterize a small molecule inhibitor for a tetrameric water channel membrane protein. Show less

      As a Research Fellow I have gained more experience in data acquisition and data analysis in solid-state NMR. I am involved in preparing samples and performing routine NMR experiments to study the structure and functions of different proteins. i have used Transmission Electron Micorscopy to study the mechansim of amyloid fibril disaggregation by chaperones. I am incharge of laboratory purchases and safety management.

      I worked on the structural and biophysical characterization of various bromodomains, including ATAD2/B and BRPF1 proteins, using my NMR expertise to identify and characterize the binding sites of mono- and diacetylated histone ligands. New skills acquired included ITC and X-ray crystal screen setup and data analysis for solving high resolution crystal structure of bromodomains in complex with histone ligands.

      My focus is on structural and functional characterization of bromodomain-containing proteins and how they select for specific histone modifications. New Skills acquired during this time include mammalian cell culture, RT-qPCR, immunoblotting. Hands on training in Cryo-EM and received Merit badges in Vitrobot use for cryo grid preparation, clipping of cryo grids and screening using Glacios microscope. Currently being trained in use of Leginon and CryoSpark softwares for data analysis.
