      Expression, purification and characterization of recombinant α-synuclein proteins:Recombinant α-synuclein proteins were expressed using BL21 (DE3) pLysS bacterial expression system under the control of T7 promoter. Bacterial cells overexpressing recombinant proteins were disrupted using ultrasonication and the proteins were purified using ion-exchange chromatography (mono Q ion exchange column) followed by size-exclusion chromatography (shodex 803) to achieve high purity. Recombinant α-synuclein proteins were characterized for their in vitro fibril formation using Fluorescence assay, Circular Dichroism (CD) and Transmission Electron Microscopy (TEM).Expression and purification of Tryptophan synthase alpha subunit mutants for structural characterization.
