  Le Champ du Ver Luisant est une petite entreprise de maraîchage biologique en traction animale.Ce stage était non seulement un stage ouvrier qui consistait à découvrir le milieu et apprendre les différentes activités, mais aussi un stage de recherche et d'analyse de l'entreprise et de son fonctionnement.

  Le stage consistait à développer un test immuno-chromatographique par détection magnétique, grâce à la migration de nanoparticules magnétiques et à des capteurs magnétiques GMR. J'ai pu ainsi mener tout les tests préliminaires pour évaluer la faisabilité du projet : développement de bandelettes immuno-chromatographiques et migration des particules, simulations informatiques codées en Python, développement d'un appareil de mesure magnétique fiable.

  Research internship in enzymology & structural biology

  > Biochemical and structural characterization of a multimodular hemicellulaseDeveloping renewable energy and products is currently a key challenge. Lignocellulosic biomass composed of cellulose, hemicellulose and lignin appears to be a sustainable solution to produce green energy. Although degradation and bioconversion of cellulose are controlled at an industrial scale, these processes could be improved because hemicellulose and lignin are still barriers that limit access to substrate. Consequently, research focus on the improvement of hemicellulose degradation and conversion. One strategy consists in discovering and characterizing new enzymes more efficient on the diverse hemicellulose structure. In a previous work, a metagenomic study on the termite microiota of P. miliatris has been conducted and led to the discovery of two homologous multimodular hemicellulases. They are composed of a glycosyl hydrolase module GH3 hydrolyzing Beta-linkage between glucosides, a carbohydrate esterase module CE1 and a carbohydrate-binding module CBM48. During this internship, I performed a biochemical and structural characterization of this protein. The productions of the protein, its modules and mutants of them have been optimized and their activity characterized. Moreover, the structure of the module CBM48-CE1 was obtained and resolved at 1,5 Å.Technics used : -Expression, production and purification of enzymes ; directed mutagenesis-Structural study : cristallography, DSF-Fonctional study : enzyme activity assay, affinity gel electrophoresis, thin-layer chromatography Show less
