  Focused on the working mechanism of several vaccine adjuvants by studying the innate immune system 72 hours post injection of mice quadriceps muscles with selected adjuvants. Studying the influence of adjuvants on the innate immune system, allows for a better understanding of the outcome of the adaptive immune response. This allows vaccines to be constructed to target specific pathogens. Our main focus was to study: 1) cellular influx and 2) gene expression profiles both locally and systemically. Show less

  Inflammatory caspases are a functional and phylogenetical group of caspases which are involved in many different inflammatory processes. In recent studies, non-canonical inflammasome activation of mouse caspase-11 and its human orthologues caspase-4 and caspase-5 were identified as cytosolic receptors of Gram-negative bacterial lipopolysaccharide (LPS). Only intracellular LPS triggered caspase 4/5/11 activation through the lipid A structure of LPS. N. meningitidis contains a specific form of LPS that is extremely potent in activating TLR4 and is reported to be too toxic for therapeutic applications. Therefore the aim was to discover how engineered LPS derivatives are recognized in the cytosol by caspase 11/4/5 and thereby activate the non-canonical inflammasome pathway. Show less
