Molecular approach of the phylogeny of Stomiidae, a family of bioluminescent mesopelagic fishesWhile marine bioluminescent organisms commonly produce blue-green light, three genera of the Stomiidae family are able to emit far red fluorescent light. In order to understand its molecular basis and appearence in evolution, it is necessary to brighten the phylogenetic position of these mesopelagic fishes. • optimisation of DNA extraction, amplification, purification and sequencing methods• comparison of various techniques for purification of PCR products Show less

Contribution to the descriptive and functional study of the bioluminescence of Ophiocomina nigraAccording to recent observations, the black brittle-star Ophiocomina nigra (ABILDGAARD, 1789) should be able to emit a mucus of weak luminous intensity. This pattern has never been described among Ophiuroidea although the number of known bioluminescent species has doubled during the 15 last years. This controlled emission of visible light should provide an advantage against visual predators. Many strategies have been suggested but very few has been demonstrated. The aim of this internship was to explore the bioluminescence of Ophiocomina nigra, precising how this ability could be helpful when she face with a predator, in this study the green crab Carcinus maenas (LINNAEUS, 1758). In addition to the description of the bioluminescence pattern of Ophiocomina nigra, first ethologic results are promising. Prey-predator interactions indicate that the brittle-star suffer about a predation rate weaker by night than by day with sight crabs. In revenge, nocturnal and diurnal predator rates are quite similar with blind crabs. This proves that she possess visual defenses against predation. Mechanical stimulation of the brittle-star increase his luminous potential. The mucus involved in this phenomenon also should be the support of a chemical defense. Its excretion makes the brittle-star unpalatable for the predator, as edibility tests show with brittle-star's arms beforehand stressed. The combination of these two properties - chemical defense and luminous signal - suggest an aposematic use of bioluminescence that it will be interesting to investigate in the future.• collection and maintenance of benthic organisms• anesthetisation, dissection, extraction of mucus• measurement of light emitting capabilities by luminometry• test of palatability, behavioural observations Show less
