Title: “Structure-Function Relationship of Human Plasma Gelsolin and Its Mutants"Global conformational changes associated full length plasma gelsolin under calcium free condition using small-angle X-ray scattering. I used small-angle X-ray scattering to show that how lowering the pH causes global conformational changes in full length gelsolin to attain an open functionally active structure from closed inactive structure. I made different truncated version of full-length plasma gelsolin and tested their ability to depolymerize F-actin in the presence of Ca2+/low pH. I demonstrated that the smallest gelsolin, 28-161, is capable of severing F-actin much better than full length protein and any other truncated version by in vitro actin depolymerization assay Show less

Understanding how G-protein coupled receptors (GPCRs) and GTPase-activating proteins (GAPs) coordinate to balance signal speed and amplitudeI worked on GPCRs and G-protein interactions. Although the basic events involved in G protein signaling are well known, but their quantitative regulation is poorly understood. To elucidate the kinetics of basic events in G protein signaling, I developed a fluorescence resonance energy transfer (FRET) based biosensor to directly monitor individual steps in the receptor regulated GTPase cycle in an in vitro reconstituted system Show less
