      Project: Study of physio-pathological mechanisms implicated in the selectivity of impairment observed in Limb-Girdle Muscular Dystrophy 2 (LGMD2)Supervisors: Dr. Evelyne Gicquel and Dr. Isabelle RichardSummary: LGMD2 is a heterogeneous group of muscle diseases, caused by the mutation in different genes that are important in the muscle physiology. Despite its various causes, LGMD2 patients have something in common: some muscles are more prone to be affected by the disease while some are more protected. Several transcriptomic profiling comparing those two opposite groups of muscles from non-human primates revealed 3 protein-coding genes and 2 miRNAs that are differentially expressed. During the 6-month internship, I had been learning how important these candidates in the muscle and muscular dystrophies. Show less

      I lead several projects with the aim to design a new class of AAV virus for fundamental research and therapeutic purposes.- Apply protein design tools to design tissue/cell-targeted AAV ¤ Inventor of novel myotropic AAV LICA1, which showed state-of-the-art muscle targeting and liver detargeting ¤ Recipient of POCUP grant (100k Euros) to develope 2nd generation of LICA1 vector, with focus on immune response and better targeting- Build giant AAV library and AAV language-based model ¤ Inventor of AI model that designs AAVs on multiple properties by modifying multiple VR sequences. Show less I participated in four investigations to uncover pathological aspects of Duchene Muscular Dystrophy (DMD), and propose novel treatment perspectives.Study 1: miR-379 links glucocorticoid treatment with mitochondrial response in DMDStudy 2: Cholesterol metabolism is a potential therapeutic target in DMDStudy 3: The DLK1-DIO3 cluster miRNAs regulate mitochondrial functions in the dystrophic muscle in DMDStudy 4: Combined AAV gene transfer with simvastatin treatment in DMD I was 1st author of 3 publications. I also supervised 3 Master students. My knowledge and skills in mitochondria, bioinformatics, miRNAs, etc., were also applied in other unrelated studies in our group. Show less
