During my time in the labs I familiarized myself with the environment and conducted various experiments in the field of cell death, in order to gain better understandings on the recently identified mode of programmed cell death, termed "necroptosis". During this project, four thermo-tolerant bioactives were evaluated in order to determine their suitability as alternatives for antibiotics in the treatment of bovine mastitis. They were assessed for their antimicrobial capabilities as well as their suitability for polymer incorporation through hot-melt extrusion (HME), which greatly opens their potential applications and uses. Well established methods in the evaluation of antibiotics and other antimicrobial compounds were utilised to allow comparisons with published literature and already well-established antimicrobials. Novel methods were developed and implemented to assess the antibiofilm capabilities of the bioactives, in order to determine the bioactives effects upon bacterial biofilm formations. Methods were also developed from pharmaceutical studies used in determining the combinational potential of the bioactives in terms of synergy scores, which were utilised to determine the most effective antimicrobial combinations of each bioactive. In tandem with antimicrobial studies, the bioactives were assessed for their cytoxicological and immunological effects upon mammallian tissue, which was determined by use of cell viability assessments and PCR analysis.A number of articles have been published with this reserach.
