Project 1: Investigating root causes of reduced API dissolution of tablet formulationKepner-Tregoe analysis was performed to break down the complexities of the problem. The effects of operation process parameters and raw material attributes were analysed. Scatter plots, contour plots and multivariate data analysis study identified the root cause of the problem. Partial least square regression model was built to predict the API dissolution for future tablet batches. Project 2: Investigating root causes behind nip zone plugging issue during roller compaction processFlow properties of API powders were measured using a powder rheometer. The study identified that the cohesiveness of API powders was one of the root causes of the problem.Received “Award of Excellence” for excellent contributions and assistance to the company in short time period. Leading a technical team of 8 process engineering & PAT scientists in Process Engineering & Analytics, Drug Product Development dept, to deliver steriles portfolio (7+ long acting injectables, 1 oligonucleotide) with following responsibilities.1) develop robust manufacturing processes and platforms for the pipeline portfolio spanning phase I to phase III and launch2) work closely with formulators, analytical scientists, materials scientists in the CMC drug product development journey and deliver assets to their respective milestones3) develop and deploy PAT tools and strategy for this portfolio4) implement predictive approaches to accelerate product development5) enhance and integrate the fundamental understanding of materials science to manufacturing process and performance.Current process/platform and PAT focus of the team: wet bead milling, lyophilization, solid-liquid & liquid-liquid mixing, filtration, filling, PAT for these processes- Process Engineering & Analytics Lead for a late-stage oligonucleotide asset – selected and implemented a commercial non-platform mixing technology for this challenging asset, deploying online Raman and pH PAT for the asset.- Leading active GSK collaborations with external industrial and academic groups in the areas of process development, formulation and modeling.Note: Fellows program across the company was terminated in Feb 2024. Show less - Steriles and Biopharm Drug Product Modeling Lead (cross-functional matrix team) with an accountability to embed Digital Design (data-driven, hybrid and mechanistic modeling) to accelerate & transform product development (May’22 - Dec’22)- Led a cross functional strategy of materials characterization workflow (MCW) for parenteral LAIs. Conceived and employed API sparing predictive tools to identify API processability and accelerate product development. Infused “predict-first” collaborative culture in daily project executions.- Industrial Advisory Board Member, representing GSK in CIMSEPP (10+ Industry & 2 University research center) (May’22-Jan’24) Show less - Process Engineering & Analytics Lead for 4 oral solid & 2 parenteral LAIs spanning phase I to III. Delivered and implemented drug product processes (roller compaction, blending, continuous & batch direct compression, wet bead milling, solid-liquid mixing) for successful formulation, process development, scale-up and tech transfer.- Developed roller compaction process at GSK. Established, validated and executed predictive mechanistic models and materials analysis. Scaled up the process at CMO site. As a team, we supplied Phase-III products for patients ahead of schedule.- Developed a viable and scalable wet bead milling processes and evaluated formulation attributes for a parenteral suspension in 1-week timeline for GLP manufacturing, reduced process time by >50%, defined phase-I process 6 months early.- Presented accomplished work in technical project reviews, town hall meetings, engagement sessions and CMC/MPD meetings. Coauthored IND, IMPD, NDA submissions and technical reports.- Developed, led & implemented a cross functional strategy – Materials Characterization Workflow (MCW) to all oral solids. MCW enabled rapid & early manufacturability assessment with <50 g API, empowered optimization of API/formulation properties, crystallization processes and use of simpler manufacturing platforms early in lifecycle.- Led the matrix “Flow” work stream, member of “Segregation” and “Implementation” work streams. Developed advanced characterization tools, applied new ways of working in flow, segregation & compaction fields.- Led active GSK collaborations with external industrial and academic groups in materials Science (segregation, flow), process development, formulation and modeling fields.
