      The terpene class of molecules were evaluated for their potential to reduce inflammation and modulate bacterial growth. Human microbiota were exposed to terpenes and monitored to assess their suitability for future experiments in which genetic engineering would facilitate the creation of microbes that could synthesize terpenes at therapeutic concentrations. These efforts promoted an increased understanding of terpene bioactivity and commensal bacteria behavior that furthered efforts toward a novel treatment for dermatological conditions. Show less Evaluated the mechanism of inositol 1,4,5-trisphosphate receptor-binding protein (IRBIT) in the regulation of intracellular calcium signaling. A CRISPR-Cas9 mediated knock-in was used to alter serine-68 of IRBIT to alanine, thereby inhibiting a phosphorylation event that has been shown to be necessary for activation. Live-cell confocal imaging assays were developed with fluorescent calcium-sensitive dyes to analyze calcium signals in wild-type and knock-in human embryonic kidney cells (HEK293). These efforts developed the methods and cell constructs needed to further understand the misregulation of calcium signaling in diseases like Alzheimer's, Parkinson's and Huntington's. Show less
