      I worked in the Chernoff lab researching the temporal and spatial variations in macroinvertebrates along the Connecticut and Coginchaug Rivers as a bioindicator of ecosystem health.

      The Health of the Outer Reef

      I examined the health of the outer reef off the coast of Ushango, Tanzania by looking at environmental factors as well as bio-indicator species such as sea urchins and algal cover. I used environmental factors such as turbidity and observed fishing habits to infer possible causes of reef damage.

      Dana Dolinoy Laboratory, Department of Environmental Health Sciences:• Identified stable alterations in 5-methylcytosine (5mC) and 5hmC at imprinted loci in the adult mouse brain after perinatal bisphenol-A (BPA) exposure. • Designed and optimized pyrosequencing assays using bisulfite/oxidative bisulfite conversion to measure 5mC/5hmC and to validate epigenetic biomarkers of in utero BPA exposure in the adult mouse brain.

      Brian Chorley Laboratory, U.S. EPA Office of Research and Development• Characterized genome-wide methylation profiles in frozen and archived formalin fixed paraffin embedded (FFPE) adult male rat liver samples after phenobarbital exposure using reduced representative bisulfite sequencing (RRBS) on the Illumina NextSeq500 platform.• Developed a bioinformatics pipeline using Python and R programming to validate epigenomic signatures generated from archived FFPE samples by comparing them with signatures generated using frozen tissue.       Joseph Bressler and Hernando Lopez Bertoni Laboratory, Department of Environmental Health & Engineering Toxicology, Physiology and Molecular Mechanisms Program • Implemented research identifying epigenetic regulation of the DNA Damage Response and chromatin architecture as targetable mechanisms for enhancing chemo- and radiosensitivity in glioblastoma stem-like neurosphere cell lines.• Designed and executed luminescence and fluorescence plate-based cytotoxicity and immunocytochemistry assays to establish enhanced efficacy of adjunctive ascorbic acid therapy with chemo- and radiotherapeutics and establish dose- and time-response curves.• Developed multiparametric cell-based assays (cell proliferation, viability, apoptosis, migration, mitotoxicity, DNA damage, and protein interaction assays) using immunocytochemistry and fluorescence imaging techniques to assess drug efficacy and elucidate mechanisms of action in patient-derived cellular models. • Established knockdown cell models using liposome-mediated lentiviral transduction techniques, collaborated with the Newby Lab to develop CRISPR/Cas9 knock-out cell models, and optimized high throughput genetic, fluorescence microscopy, and flow cytometry-based screening protocols to identify successfully edited clones.• Analyzed existing literature and publicly available glioma patient RNA-seq and bisulfite-seq datasets (CGGA, TCGA) to generate hypotheses for potential prognostic biomarkers, therapeutic targets, and disease-relevant cellular readouts for glioblastoma treatment resistance and tumor recurrence.
