  Senior Sophister Research Project

  "Investigating the role of LspA in virulence and antibiotic resistance in Staphylococcus aureus"As part of my senior sophister year in Microbiology, I completed an individual 9 week research project under the supervision of Professor Joan Geoghegan. This research involved inactivation of the signal peptidase II gene (lspA) of Staphylococcus aureus in order to determine its potential influence on specific virulence factors of the bacterium, and thus explore its suitability as a target for anti-staphylococcal therapy. Furthermore, given the increasing emergence of antibiotic resistance, the role of LspA in mediating antibiotic resistance was also examined, specifically resistance to oxacillin and vancomycin in the methicillin-resistant USA300 strain LAC.Techniques I used during my project included:- Aseptic technique- Bacteriophage transduction- Growth curves- Solid phase adhesion assays- SDS-PAGE & Western Blotting- Agarose gel electrophoresis- Red blood cell lysis assays- MIC assays- PCR- Molecular cloning- Preparation of competent cells and electroporation.
