      The conducted PhD thesis explores the introduction of artificial amino acids into an entire zinc finger protein to incorporate a cleavage tool into a DNA binding biomolecule. The two main synthetic strategies used were Fmoc solid phase peptide synthesis (SPPS) and protein expression to achieve this goal. The sequence which encodes two zinc fingers (60aa) was cloned into a specific vector releasing the protein as C-terminal thioester during purification. The two main synthetic strategies used were Fmoc solid phase peptide synthesis (SPPS) and protein expression to achieve this goal. The sequence which encodes two zinc fingers (60aa) was cloned into a specific vector releasing the protein as C-terminal thioester during purification. A set of peptides were synthesised composed of the third zinc finger (30aa) with an N-terminal cysteine and several different artifical amino acids included into the sequence. Subsequent ligation lead to 6 different poly peptides which were analysed by ESI mass spectrometry and CD spectroscopy. Also organic synthesis was used to prepare artificial amino acid building blocks to be incorporated in additional poly peptides. Show less

      My Bachelor thesis in 'Forensic and Analytical Chemistry' entitled 'Extraction of DNA from bone' dealt with the question how we can enhance the extraction of DNA from bone material. For that purpose PCR and capillary electrophoresis were used to compare several commercially available extraction kits.

      Activities and Societies: Student Union Organic synthesis of artificial amino acid building blocks for solid phase peptide synthesis (Fmoc). Metal binding bisimidazole moieties were introduced into the side chain of natural amino acids in order to create novel metal binding sites in zinc finger protein Zif268, a DNA binding transcription factor.

      I have been involved in several projects at the interface of chemistry and biology using my expertise in peptide chemistry and protein ligation to support several teams. I have been also involved in a project aiming to semi-synthesise short homogeneously glycosilated proteins by chemically modifying recombinant expressed proteins.As intern of the Business Development Course 'Molecules to Medicine' and as part of the 'on-the-job-training' I have developed a business plan for a project in a different scientific area. This experience has provided me with a new skill set and sparked my interest to explore the business side of projects. Show less

Based on the completed detailed competitive analysis and path to market for the technology the internal business development unit at CSIRO assessed the commercial viability of the project and steps are taken to pursue this goal.
