  "Confimation and characterization of a putative target of the U-box type E3 ubiquitin ligase PUB24"In my current project, I employ in vivo and in vitro protein-protein interaction techniques to assess PUB24 interaction with one of its putative target proteins, which is known to be crucial for the regulation of vesicular trafficking. Furthermore, I investigate the potential target ubiquitination by the action of PUB24 and its implication in the regulation of Arabidopsis immune response.Relevant Skills Acquired: Protein-protein interaction assays: in vitro pull-down, MST; in vivo Co-IP, BiFC, split-luciferase assay Show less "Phosphorylation of the E3 ubiquitin ligase PUB22 controls its ubiquitination activity to dampen the immune response"In my Ph.D. project I discovered a novel molecular mechanisms controlling E3 activity and the first to be reported in plants. PUB22 is regulated by autoubiquitination, which results in its constant degradation. MPK3-mediated phosphorylation stabilizes PUB22 by reducing its autoubiquitination levels. Autoubiquitination is dependent on the oligomerization of PUB22, which is inhibited by phosphorylation. MPK3-mediated phosphorylation of PUB22 triggers an activity switch by disrupting oligomerization, rescuing PUB22 from degradation and allowing it to engage targets to dampen cellular signalling during immunity.
