      I conducted an in vitro cell culture study to examine the role of myeloperoxidase (MPO) induced oxidative stress in isolated human umbilical vein endothelial cells (HUVECs). Obtained skills: - Isolation of HUVECs - Cell culture- Sample preperation for Mass Spectrometry- Fluorescent Assays

      This research project examined the influence of systemic inflammation on fracture healing. We studied whether systemic inflammation in multitrauma patients leads to impaired fracture healing by conducting an in vitro proliferation and (osteogenic) differentiation study with human multipotent stromal cells (MSCs) using single cell analysis techniques.Obtained skills:- Cell culture of MSCs- FACSThis internship included clinical experience in the surgery department, in which I was trained to perform venipunctures and assisted in trauma surgeries.       In reconstructive surgery, fat grafting is used as soft tissue filler for soft tissue defects in burn wound healing. However, due to resorption and volume loss of the injected fat, the long-term fat graft longevity has shown to be inconsistent and unpredictable. Therefore, I performed an in vitro cell culture study to examine the possible benificial effect of activated platelet rich plasma (PRP) on human fat fibroblast like cells (hFLCs). Cell activity, proliferation and differentiation of hFLCs were cultured with PRP; the co-cultures were analysed by the conduction of MTT assays, BrdU labeling and immunohistochemistry.Obtained skills:- Isolation of human fat fibroblast like cells- Cell culture- Immunohistochemistry- Fluorescent assaysThis internship included clinical experience in the department of plastic, reconstructive and hand surgery, in which I assisted in surgeries and medical procedures.       The hippocampus harbors neural stem cells (NSCs) and neural progenitor cells (NPCs), and a microenvironment that supports adult neurogenesis; hippocampal NSCs are located in the subgranular zone (SGZ) of the dentate gyrus,and give rise to new functional and integrated neurons in the hippocampus throughout life. During this research project, the role of microglia in adult hippocampal neurogenesis was evaluated in an in-vivo study in mice. This study revealed the effect of aging and microglia on proliferation, fate and survival of Neural Progenitor Cells (NPCs). These processes of neurogenesis were determined by BrdU labelling of NPCs, immunohistochemistry, and by confocal microscopy analysis.Skills obtained:- Laboratory animal work- Imunnohistochemistry- Confocal microscopy Show less

      During my graduation internship, we conducted an in-vitro and in-vivo study on the remodelling of cerebral arteries after subarachnoid hemorrhage. The study was designed to characterize in vivo and in-vitro vascular wall remodelling of cerebral arteries following subarachnoid hemorrhage in rats and mice. Contractile properties of cerebral arteries were determined and immunohistochemistral techniques and confocal microscopy were used to analyze brain tissue.Obtained skills:- SAH model in rats- Wire myograph- Immunohistochemistry- Confocal microscopyThe internship included obtaining clinical experience in the Neurology department. The theme of the congress was “The Walk of Life“, in which the use of different technical innovations in the diagnosis and treatment of patients in different stages of life were highlighted.
