  "Molecular targeting of proteins by homocysteine: implications in familial and clinical hypercholesterolemia""S-homocysteinylation (formation of disulfide bounds between homocysteine – Hcy thiol group and cysteine residues of proteins) and N-homocysteinylation (acylation of free amino groups mediated by the toxic metabolite of Hcy, Hcy-thiolactone – HcyTL) offer a new paradigm to be considered in the vascular pathogenesis of hypercholesterolemia (1). These post-translational protein modifications have been implicated in protein inactivation, oxidative stress and reverse cholesterol transport dysfunction (1). The HcyTL detoxification is mediated through the lactonase (LACase) activity of paraoxonase-1 (PON1), an HDL-associated enzyme, avoiding protein N-homocysteinylation (2). Aim: To compare protein S- and N-homocysteinylation between normocholesterolemic (NC), and both clinical (CH) and familial hypercholesterolemic (FH) groups."What I learned in the context of this project:-colorimetric assays of paraoxonase activities on 96-well plates;-analyte quantification by fluorescence and UV-HPLC;-biological samples manipulation (blood and serum);-use of statistic program (Graphpad) for data analysis;-working with large clinical databases.
