  Thesis: “Application of biomolecular techniques for the microbiological characterization of sites contaminated by chlorinated solvents" The work carried out in this thesis has been focused on the molecular characterization of a microbial community, in particular of microorganisms involved in reductive dechlorination processes that are naturally resident in an aquifer contaminated by chlorinated solvents.The aim of the work was to contribute to the clarification of the role and the interconnections between the various organisms present in the analyzed ecosystem and to improve scientific tools for the development of intervention strategies for the recovery of polluted groundwater from these compounds.The monitoring of the microbial community was carried out in order to define the level structure of the main bacterial phylaWe exploited a biomolecular technique of "in situ hybridization", the CARD-FISH (Catalized Reporter Deposition-Fluorescence In Situ Hybridization), together with direct microscopic analysis and cell counting. By using the same methodology, we also monitored specific dechlorant microorganisms, such as bacteria belonging to the genus Dehalococcoides spp., to date the only one known to be able to support the complete reductive dechlorination to non-toxic ethylene. We concluded that these microorganisms can be used as bio-indicators of the site contamination and their detection in environmental samples can provide important information about the potential of bioremediation of the contaminated site. Furthermore, the CARD-FISH proved to be a powerful technique that thanks to a culture-independent approach makes possible to study microorganisms of interest as they appear in an environmental matrix, that is the more complex indigenous microbial communities.
