      Characterization of the active site of The Escherichia coli enzymes cystathionine gamma-synthase (eCGS). The e.Coli enzymes cystathionine gamma-synthase (eCGS) and cystathionine beta-lyase (eCBL) are almost identical in overall structure, with a root mean square deviation for superposition of the carbon alpha atoms of the peptide backbone of only 1.5 Å. While many active-site residues are conserved between eCBL and eCGS, a notable difference is a pair of aromatic residues (F55 and Y338) of eCBL that are replaced by the acidic amino acids D45 and E325 in eCGS. Previous work from our lab and comparison of the structures of these 2 enzymes suggested a potential role in substrate binding and/or regulation of reaction specificity. A series of 12 site-directed variants of these 2 positions (F55A, F55E, F55H, F55K, F55L, F55N, Y338A, Y338E, Y338H, Y338K, Y338L and Y338N) were constructed to explore their roles in the active site of eCBL in greater detail. These variants were expressed in E. coli host cells and purified using nickel nitrilotriacetic acid affinity chromatography and concentrated for future kinetic assays. Show less

Steady state kinetic parameters conducted to compared the wild-type enzyme to mutants in order to determine the effect of each substitution on substrate binding and catalysis.
