      Recombinant antibody for diagnosis of hepatitis B- Designed the primer for ScFv (single-chain fragment variable) an anti-HBsAg and amplified the target region by PCR, subcloned the recombinant ScFv in pGEM-T Easy vector through T/A cloning and confirmed the sequence by restriction digestion and DNA sequencing. - Expressed the protein in a bacterial expression system, optimized the induction time and growth condition for protein expression.- Purified the protein using affinity chromatography and performed functional characterization by ELISA.       My PhD work focused on:- investigating the effects of hepatitis C virus (HCV) core protein on intracellular signalling pathway in human T cells and unravelling the molecular mechanism of core protein-mediated epigenetic modification of SHP-1 promoter, a tumor suppressor gene and a negative regulator of immune signalling pathway by bisulfite sequencing.- establishing a unique protocol for NGS based sequencing library generation by using Illumina specific adapters and a dual barcoding system for profiling of DNA methylation in cell models and PBMC samples from blood donors and HCV patients.- exploring the mechanism of calcium release in human T cells by HCV core protein to understand the viral pathogenesis and HCV life cycle.- establishing a PCR based method for the separate detection of two strands of HCV RNA to monitor virus replication and track active infection.       During the pandemic season 2020-2021, when the clinic was in urgent need of rapid development of novel diagnostic techniques and skilled technicians, I took a temporary leave from my PhD studies and volunteered to work at Akademiska´s diagnostic laboratory in Uppsala. Assisted senior microbiologist Dr. Anders Bergqvist with the development and optimization of a PCR-based typing method for screening of several SARS-COV-2 variants, B.I.I.7/alpha, B.I.351/beta, PI/gamma and B.617.2/delta. To provide the contact tracing and surveillance, I volunteered to assist with testing the patient samples to screen the variant of concern. The role involved picking and storing the positive samples, performing the typing PCR and data analysis. Show less
