      • Induced transformation of mutants of Streptomyces lividans endoglucanase CelB2 into E.Coli strain BL21 using the QIAGEN test prep protocol.• Conducted a multi-step purification process through the use of procedures including Amylose Affinity Chromatography Columns and DEAE Anion-Exchange Chromatography Columns.• Analyzed elutions between purification steps using 1% - 7% gel electrophoresis and spectrophotometry .• Carried out enzyme activity assay to determine specific activity, yield %, and purification fold of protein.• Resulted an increase in purification of protein by 64 fold. Show less • Used the sensitive biomarker assay EROD (enzyme 7-ethoxy- resorufin-O-deethylase) to test induction for two genes correlated with toxicological endpoints of the Zebrafish model (cytochrome p450 producer gene CYP1a and transcriptional repressor gene TCF3a).• Used RT-qPCR to isolate RNA and observe induced expression for both genes. • Tested Triclosan as a Mutagenic compound using the ‘E.coli Mutagenicity Test Kit’ protocol through observing reverse mutations caused by the chemical in question.• Used C-18 reverse phase HPLC (High Performance Liquid Chromatography) column as a separation technique to test for the formation of triclosan metabolites (toxic and non-toxic) when reacted with S9 hepatic enzymes (cytochrome P450 and other conjugates).
