Activities and Societies: Master in Philosophy in Biotechnology major. I worked on dissertation entitled"Liposome based formulations of polyene antibiotics in the treatment of infections." My M Phil work was focused on development and surface modification of liposomal based formulations of polyene antibiotics. Polyene antibiotics are used against systemic infections and possess strong activity against infections like C. albicans and C. neoformans. But these antibiotics are associated with high hemolytic activity when used at high dosage. I developed phosphatidyl choline and cholesterol based liposome formulations. But these antibiotics are associated with high hemolytic activity when used at high dosage. I developed phosphatidyl choline and cholesterol based liposome formulations. We did their surface modification using tuftsin. Tuftsin, is a immunomodulator and it boost immune cells when come in contact. Combination of tuftsin with liposomes encapsulating these antibiotics, enhances their efficiency in combating infections. These formulations were examined for in vitro toxicity and are shown to be non-toxic even when used at high dosage of encapsulated antibiotics. Further, they were found highly effective and non-toxic when used again systemic infections like Candidiasis and Cryptococcis in mice model. Show less

I developed poly-lactic glycol acid (PLGA) based nano particles of perillyl alcohol (POH), to encounter poor pharmakokinetics and pharmakodynamics associated with POH. I studied their efficacy in di methyl benzo anthracene (DMBA) induced skin papilloma. Developed nano particles were studied for their size and release kinetics of POH using HPLC. We studied reduction in tumor size, apoptosis using Western blot of Caspases, and survival of treated animals. Since PLGA nano particles were associated with burst release pattern, we tried to encapsulate them further in phosphatidyl choline (PC) liposomes to overcome burst release. This strategy allowed us to pass burst release pattern. Nano formulation encapsulating curcuming proven to be highly efficient in delivering it at slow rate. We tested its efficacy against hepatocellular carcinoma and they were found to suppress carcinoma. Show less

I worked on two different projects at Molecular connections. One project Immunex was focused on curating the immunological response data associated in different diseased conditions. Another project Proteome was focused on putting together the scientific literature associated with proteins that plays a major role in disease pathogenicity.

My postdoctoral expertise added advancement to my skills. I workedon PTEN (phosphatase and tensin homolog), a tumor suppressor gene. I generated endothelial specific PTEN deleted mice. I confirmed the PTEN deletion via quantitative PCR and Western blotting of PTEN. Further I examined role of PTEN deletion in the mouse model. To address its role in acute lung injury, I used LPS-induced sepsis models. I demonstrated that the PTEN knock-out mice have a prolonged life span and they are resistant to sepsis. Further I studied lung endothelium integrity by means of Evans blue dye where I injected albumin-conjugated Evans blue retro-orbitally and measured its leak into lungs. I established that PTEN-deficient mice had protected lung endothelium. To explore the signalling mechanism responsible for this function, I studied Akt mediated signalling and cell proliferation using flow cytometry techniques. I also worked on several other projects with my colleagues focused on signalling in lung endothelium during sepsis. Show less

As a part of nanotechnology institute, I am trying to develop nanoemulsion based vaccine for mucosal immunization. I modified the nanoemuslion in a way that could boost the immune responses mucosal surface and helps in successful elimination of the pathogen. Currently, I am working on a nanoemulsion and its modification that could boost antibody responses as well as provides balanced Th1/Th2/Th17 responses for mucosal immunization.
