We recently identified a novel interaction between nonstructural protein 15 (NSP15) and cytokeratin by mass spectrometry and confirmed this interaction by immunoprecipitation. As cytokeratin is characteristically expressed in epithelial cells, which are the primary target cells of SARS-CoV-2, it will be interesting to see how this interaction between NSP15 and keratin alters the cell's cytoskeleton to promote viral proliferation.       The focus of my Master's thesis is on exploring protein-protein interactions between host proteins and SARS-CoV-2 nonstructural proteins (NSP) to elucidate the function of these viral proteins. As part of my thesis I successfully cloned 3 of the 16 NSPs into pET28 plasmids, expressed them in BL21 E. coli and identified a novel interaction between NSP15 and keratin. I then validated this protein-protein interaction by immunoprecipitation and Western blot. I followed up by transfecting the viral protein into Vero cells to demonstrate that the presence of NSP15 leads to alterations in the keratin (IF) cytoskeleton and causes cell death via immunostaining and an MTT colorimetric assay respectively.Through these sets of experiments I was able to discover a novel protein-protein interaction between NSP15 and cytokeratin-1 and also demonstrated a potential novel mechanism of action for how this viral protein promotes the viral lifecycle during infection.
