Purification and Characterization of Challenging Proteins• Designed and constructed >100 recombinant protein expression vectors with desired structural modifications.• Developed the purification and refolding process to produce challenging proteins in their biologically active forms, and used as therapeutic targets.• Mastered in versatile protein purification and characterization methods including FPLC AKTA systems, SDS-PAGE, western blot, mass spectrometry and binding assays (SPR, FP, ELISA) to deliver 50+ high-quality proteins.II. Discovering and Evaluating Cell-Penetrating Peptides (CPPs)• Designed and synthesized a novel group of highly active disulfide-cyclized cell-penetrating peptides.• Established the complete structure-activity relationship (SAR) profiles of new cell-penetrating peptides.• Demonstrated the potency of discovered peptide by successful intracellular delivery of protein cargoes.III. Developing Peptidyl Inhibitors against Protein-Protein Interactions• Discovered potential peptide inhibitors against immune checkpoint protein-protein interactions through library screening.• Optimized the synthetic strategy of a challenging peptide ligand and applied for collaborative animal studies.IV. CPP-Protein Conjugate Analytics and Characterization• Manufactured 10+ CPP-protein conjugates through various site-specific chemical and enzymatic ligations.• Characterized the conjugation efficiency, stoichiometry, and stability of protein conjugates via different analytical approaches.• Demonstrated the efficacy of CPP-protein conjugates by successful intracellular delivery of model proteins.
