      Research in the INSERM BioMaterials & BioEngineering Laboratory focuses on developing and characterizing new generations of biomaterials and their integration in the tissue microenvironment by mimicking biomechanical cues. This PhD research was performed under Dr. Dominique Vautier & Pr. Youssef Haikel. This work led to 2 peer-reviewed publications.• Focus on the understanding of the effect of cellular microenvironment stiffness on chromatin plasticity.• Investigation of the fate of epithelial healthy and cancer cells as well as interaction at cell/materials interface.• Mechanical characterization of polyelectrolytes multilayers.• Cell culture experience with epithelial cells (PtK2, SW480, 3T3).• Use of diverse imaging techniques – Epifluorescence Microscopy (Nikon Eclipse Ti-S), Confocal Microscopy (Zeiss LSM 510), Fluorescence Recovery After Photobleaching (FRAP), Timelapse (Nikon Ti-E).• Use of highly flexible software for data analysis – FIJI/ImageJ.• Use of a wide range of analysis techniques such as Immunocytochemistry, Life/Dead assays, and Pharmaceutical Inhibition.INSERM Workshops: • Live imaging of morphogenesis in 4D• Single cell sequencing: from basic research to clinical application Show less

Postdoctoral research performed under Dr. Amit Pathak. This work will lead to a paper currently in preparation.• Focus on the effect of polyacrylamide collagen-coated gels stiffness on the nuclear architecture of epithelial breast cells.• Mechanical characterization of polyacrylamide gels.• Cell culture experience with epithelial cells (MCF10AWT, MDA-MB-231 and derived cell lines) and macrophages (RAW 264.7).• Use of diverse imaging techniques – Confocal Microscopy (ZEISS LSM 880 Confocal with/without Airyscan), Fluorescence Recovery After Photobleaching (FRAP), Timelapse (Zeiss Celldiscoverer 7, Zeiss Axio Observer 3 and Olympus IX81) and Transmission Electron Microscopy (JEOL JEM-1400 120kV TEM).• Use of highly flexible software for data analysis – FIJI/ImageJ, Imaris, MATLAB.• Use of a wide range of analysis techniques – Western Blot, Immunocytochemistry.
