Collaboration and training experience, continuation of the research activity started with the experimental Master Thesis on the CNR Searcular project with the editing of scientific publications.Acquired skills:• Dynamic Light Scattering (DLS) and Multi-Angle Dynamic Light Scattering (MADLS), use of Zetasizer Ultra Red (Malvern Panalytical);• Manipulation of eukaryotic cell cultures: proliferation assay (Hoechst 33342), collagen deposition assay in cultured 3T3-NIH fibroblasts (Sirius Red Total Collagen Detection Assay).Initial approach to:• LC-MS/MS and Top-Down approach;• Use of the following bioinformatic tools applied to proteomics: Panther, Grid, Proteome Discoverer, BIOPEP-UWM;• Construction of a plasmid vector;• Transformation of bacteria (E. coli) with plasmid DNA, expression and purification of recombinant proteins;• Phage Display technique. The work concerned the extraction of Acid Soluble Collagen from organic extracts of fish bones (Salmo salar and Sparus aurata), the biochemical characterization and the evaluation of the biological activities according to the degree of purification.Acquired skills:• Protein purification and protein characterization;• Solid Phase Extraction (SPE);• Protein quantification (Nanodrop, Bradford test and Bradford-SDS variant) and collagen quantification in organic extracts with hydroxyproline assay;• Extraction of Acid Soluble Collagen (ASC) from organic bone extracts;• SDS-PAGE protein electrophoresis and Western Blot;• HPLC and use of Xcalibur software;• UV-Vis Spectroscopy;• Use of different microplate readers;• Manipulation of eukaryotic cell cultures: maintenance, dilution and cell expansion; cytotoxicity studies and cell viability assays in cultured 3T3-NIH fibroblasts;• Sterilization and freeze-drying processes;• Evaluation of antioxidant activity: in vitro biochemical assays (ABTS, DPPH); antioxidant assays on 3T3-NIH fibroblasts by induction of oxidative stress with hydrogen peroxide;• High Resolution Respirometry (HRR) studies on intact 3T3-NIH fibroblasts (Oxygraph-2k, Oroboros);• Evaluation of antimicrobial activity (MIC) in E. coli;• Wound healing and migration assay in cultured 3T3-NIH fibroblasts (Culture-Insert 2 Well, ibidi);• Statistical data analysis (GraphPad Prism);• Communication and presentation of data and results in presence or remotely via Google Meet, Microsoft Teams, Skype and Zoom platforms.
