      The involvement of NGF and acetylcholine in mechanisms of neuronal plasticity was my main subject of interest. Several lines of evidence suggested that acetylcholine (ACh) and the neurotrophins profoundly affect neuronal plasticity in the hippocampus and neocortex. The cholinergic system is crucially involved in learning and in short- and long-term memory. Signaling by BDNF and its receptor trkB has been shown to play important roles in neuronal plasticity, both under physiological and pathophysiological conditions. However, an involvement of the prototypic neurotrophin nerve growth factor (NGF) and its receptors trkA and p75NTR in these functions has not been profoundly investigated. My job was to set up the sharp electrode and field potential recording methods in order to investigate the impact of NGF-signaling on neuronal plasticity in the amygdala and hippocampus at electrophysiological level (LTP, LTD, PPFand persistent activity).       Neurozentrum Freiburg, is one of the biggest University Medical Hospital for brain surgery in Germany, mainly for epilepsy, therefore my main project was focusing on Levetiracetam (LEV), one of the most important anti-epileptic drugs. Although binding studies have shown that LEV interacts with the synaptic vesicle protein 2A (SV2A) the mechanism of action largely remains unknown. Therefore I used hippocampus slices of juvenile rats (P17-P22) and human material, in order to study the electrophysiological effects of LEV on granule cells (GCs) and fast spiking basket cells (BCs) from dentate gyrus (DG), hippocampus. Our results are the first report showing that LEV modulate the synaptic transmission in epileptic tissue. Weniger anzeigen
