      • I found that Berberis Libanotica, a Lebanese medicinal plant, antiproliferative effects on adult T cell Leukemia.• I climbed to high altitudes to collect the plant. I classified it after detailed dissection and deposited a sample to the herbarium. • Gained a great knowledge in different extraction procedures as well as to prepare the extracts for testing them on leukemic cells.• Acquired skills for culturing cells in suspension and antiproliferative assays.       First project:• Designed and constructed ERα /ERβ chimeras (Gibson Assembly) which were successfully used shedding new light on the diverse pharmacological properties of antiestrogens.• Developed, validated and executed assays (such as FRET, BRET, western blot, pulldown assays) to monitor Estrogen Receptor and the chimeras’ interactions with SUMO and Ubiquitin in live cells. We also used Luciferase assays and qRT-PCR to monitor the activity of the chimeras in ER negative cell lines.• Discovered a novel mechanism by which of PIAS enhances ER modifications.We did some overexpression and Know downs (shRNA against E3 ligases).• Played a key role in improving the BRET technique into the future era of monitoring multimeric protein assembly in live cell (BRET FECT or Spliced BRET).Second project:• Designed and constructed ER mutants found in breast cancer metastasis resistant to endocrine therapy using site specific mutagenesis.• Uncovered a novel mechanism by which ER mutants activate transcription in the presence full antagonists and inverse agonists.• Designed a High Throughput chemical screen to measure the efficacy and potency of newly developed antiestrogen and antiandrogen HDACis hybrids.• Significant Contributions to quality data for Grant applications.• Trained Graduate students and postdoctoral fellows in mastering techniques needed fortheir projects.
