The goal of my bachelor thesis was to set up and optimize the purification of a novel protein Ivy1 from E. coli. and to identify the effect of this protein on the small GTPase Ypt7 by using SDS-PAGE, HPLC, and ITC.Our work was published as: Rai, A., Oprisko, A., Campos, J., Fu, Y., Friese, T., Itzyen, A., Goody, R., (2016). "bMERB domains are bivalent Rab8 family effectors evolved by gene duplication." Elife 5.During this project, I successfully performed: • Cultivation of E. coli and protein expression in the E. coli cells• DNA cloning by using PCR • Purification and analysis of proteins by using size exclusion chromatography (SEC), affinity chromatography, high-performance liquid chromatography (HPLC), isothermal titration calorimetry (ITC) and mass spectrometry Show less

The mTOR complex 1 (mTORC1) is a master growth regulator in mammalian cells. In my master thesis, I proposed a novel model for the amino acid-sensitive Rag GTPases cycling upstream of mTORC1 activation via verifying the regulation of the factors alone or in combination. Our work was published as: Su, M. Y., Morris, K. L., Kim, D. J., Fu, Y., Lawrence, R., Stjepanovic, G., Zoncu, R., Hurley, J. H. (2017). "Hybrid Structure of the RagA/C-Ragulator mTORC1 Activation Complex." Mol Cell 68(5): 835-846 e833.More publications are in preparing.During the time, I successfully performed a number of biochemical and molecular biological techniques:• Cultivation and infection of Insect cells and mammalian cells • MBP-, GST- or His-tagged proteins expression and purification from the different cell cultures• Plasmids design and new DNA constructs creation by using PCR• In-vitro and in-vivo analysis of guanine nucleotide hydrolysis/exchange on proteins by using the optimized methods• Transfection of mammalian cells by using RNA-Interference • In vivo- and in vitro analysis the interaction and location of the proteins by using immunofluorescence, immunoblotting, SDS-PAGE and FPLC Show less
