      FYP- "Investigating the role of ubiquitination in regulating presenilin functions in Alzheimer's disease and innate immune signalling". For this project,I performed a site directed mutagenisis to generate 2 mutant strains using 2 DNA primers to make intentional specific changes to the DNA sequence of the gene responsible for regulating presenilin proteins. For this project,I performed a site directed mutagenisis to generate 2 mutant strains using 2 DNA primers to make intentional specific changes to the DNA sequence of the gene responsible for regulating presenilin proteins. A series of methods were executed to develop a mutant strain and investigate if the manufactured mutants had any relevance to presenilin function, the steps involved; Electrophoresis, PCR, DNA purification, Digestion, Transformation, Sanger sequencing, Calcium phosphate transfection, SDS-PAGE. From this, it would have been possible to examine if the PS2 protein was expressed in western blot analysis (unsuccessful) followed by the demonstration of ubiquitin binding by GST-tagged recombinant proteins and analyse on SDS-PAGE gel and determine if ubiquitination accurs and if so the role they have in presenilin protein, specifically PS2.
