      Phd in molecular and cellular pharmacology

      My doctoral research was studying the specificity of protein tyrosine kinases and their inhibitors using structural, biochemical, and biophysical techniques. My thesis was divided into two sections: characterization of highly specific bi-substrate competitive Src inhibitors from DNA-templated macrocycles and determining the molecular mechanism of discoidin domain receptor 1 (DDR1) promiscuity for binding small molecule kinase inhibitors. For the first portion of my thesis, I crystallized and determined the structure of Src kinase domain in complex with a novel ATP- and peptide- competitive inhibitor. I also biochemically and biophysically characterized novel inhibitors to demonstrate ATP- and peptide-competiveness. To determine the molecular mechanism of discoidin domain receptor 1 (DDR1) promiscuity for binding small molecule kinase inhibitors, I established a baculoviral expression and purification system to express and purify DDR1 to biochemically and biophysically characterized DDR1’s behavior towards a variety of kinase inhibitors. I also co-crystallized the kinase domain of DDR1 in complex with several different inhibitors to better understand how DDR1 accomodates chemically different small molecule inhibitors. Show less

      My post-doctoral work was focused on the study on Wnk kinases and how they regulate the cellular response to changes in osmolarity. I utilised CRISPR/Cas9 technology to tag and knockout individual Wnk kinases in cells to generate useful tools that can be used to study the individual role of these conserved kinases in their signalling cascade. I biochemically characterized small molecule Wnk kinase inhibitors and identified potential Wnk interaction partners identified via mass spectrometry. I also helped develop protocol for the MRC-PPU to utilise CRISPR/Cas9 technology across multiple cell lines. Show less

      I was a biocurator for the UniProt knowledgebase, Open Targets, and Gene Ontology Annotation (GOA) Project. My work for UniProt mainly focused on curating the latest information on mammalian proteins so that UniProt was kept up to date on the latest findings and publications. For Open Targets, I provided ontology mappings between UniProt and the Experimental Factor Ontology that the Open Target Platform utilises. This platform is geared towards linking clinical and research data for rare diseases so new disease targets can be found. For the GOA project, I curated data using the Gene Ontology and provided assistance to the users of QuickGO, the gene ontology annotation browser, through the use of webinars and hands on training.
