      Aug 2005 - May 2006
      Student Research Technician

Observed developmental oxidative stress in the zebrafish HSP-eGFP (Danio rerio) model system. The zebrafish was genetically modified to emit green fluorescence in areas of oxidative stress under ultraviolet light. Examined the developmental toxicity using thiomersal, mercury chloride, mercury nitrate, ethanol and various types of nanoparticles such as fullerenes, multi-walled nanotubes, single-walled nanotubes and carbon chains. After dosing the embryos, they were observed using fluorescence microscopy. These genetically engineered fish fluoresced in areas of the central nervous system indicating neurological damage. A paper that came from my research entitled “Developmental Toxicity of Nanoparticles in the Zebrafish, Danio rerio” was presented at the first International Conference on Nanotoxicology: Biomedical Aspects. Aug 2006 - May 2008
      Research Laboratory Technician

Familiar with two types of human breast cancer cell lines, MCF-7 and NCI-ADR, human ovarian lymphoma cells HeLa and also murine macrophage cells J774-A1. Performed various cell viability assays with novel anti-cancer agents, mainly aza-anthrapyrazole compounds with tertiary amine side chains. Cells were then dosed at various concentrations and different time points and observed using fluorescence microscopy. Localization, distribution and excretion of the drug was determined and hypotheses were created on how cell lines work to eliminate drug and evade cancer chemotherapy. Show less

Previously conducted research in the laboratory of clinical pharmacology within the Experimental Therapeutics Program now known as the Department of Translational Hematologic and Oncologic Research (THOR) of the Taussig Cancer Institute. Previously worked with ovarian, endometrial, lung, leukemia, and renal cell carcinomas examining drug sensitivity in order to maximize efficacy and reduce toxicity of cancer chemotherapy. Currently modifying Topoisomerase enzymes and transfecting them to cells then determining the biological effect of the modification. Performing western blotting and real-time PCR analysis to assess the molecular effect of the transformation. Through modification of these topoisomerase enzymes we determine its regulatory effect during cell growth and differentiation. Through the studies with topoisomerase II, we have determined multiple defective phosphorylation sites that cancer cells use as a mechanism to evade chemotherapy. Using engineered model systems, we are learning to understand how addition of phosphate at specific sites makes this enzyme vulnerable to drug therapy in order to develop novel treatment strategies. I am part of the research team that submitted the following abstracts for the American Association of Cancer Researchers (AACR): Tyrosine phosphorylation of RIN1 modulates the anti-proliferative effects of sorafenib and sunitinib in renal cell carcinoma (RCC) (2010) and Over-expression of cisplatin resistance-associated over expression protein (CROP) is correlated with platinum resistance in ovarian cancer (2009).
