  I worked at the project of functional characterization of (unknown) mitochondrial carriers of the SLC25 family. This involves culture of yeast (S. Cerevisiae) in carbon-free medium which is supplemented with either a C12 or C13-carbon source. From this yeast culture, extraction of the metabolites will be done and analysed with a HPLC-MS metabolomics approach. When the extraction procedure of these two metabolite pools (i.e. light-labeled vs. heavy-labeled) and the HPLC-MS analysis protocols are optimized, these metabolite pools will be used in a transporter experiment. First, the known human ADP/ATP-exchanger will be expressed, purified and reconstituted with liposomes to generate proteoliposomes. Second, these proteoliposomes will be loaded with the light metabolite pool and filter-extracted, after which these loaded proteoliposomes are added to the heavy metabolite pool where transport occurs. Third, the proteoliposomes are removed from the medium (external fraction) through filter extraction and subsequently lysed (internal fraction). Last, both the internal and external fractions are measured with the optimized HPLC-MS assay to validate the complete experimental setup. Show less
