  The main task was to evaluate DNA-DSB in H4IIE cell line exposed to environmentally relevant concentrations of copper, nickel and zinc singly and in combination. In this project, agarose gel electrophoresis was used to analyse the DNA-DSB induced by heavy metals (copper, nickel, zinc) toxicity. In this method, in situ lysis of cellular material was done to reduce the mechanical damage of DNA.   Traffic is a major source of urban air pollution, an important health problem worldwide and also in Norway. Traffic pollution comprises a mixture of gases and particulate matter (PM) from exhaust and wear of road pavement, tires and brakes. From a regulatory view, the main pollution issues related to vehicle traffic in Nordic countries are high levels of coarse mineral-rich wear PM from abrasion due to the use of studded tires and exhaust PM from diesel/biodiesel vehicles. It is a major political issue to improve the urban air and the health of the population in Norway. Main tasks:- Examine the combined effects of PM from different stone types and DEP- Examine the combined effects of PM from different stone types and biodiesel- Examine signalling mechanism involved in the combined effects; with a major focus on IL-1 beta, known to be a critical pro-inflammatory mediator in inflammation- Oxidative stress in particle toxicology- MAPK/NF-kB- RT-PCR- RNA sequencing- Peer-reviewed articles and systematic review article writing Show less PrevenTAP project.Main tasks:Different PM from stone types used in road pavement was used. As combustion particles, we used MAPCEL diesel (from old cars) and B7 (diesel containing 7% biodiesel, currently used as diesel in Norway) and B20 (containing 20% biodiesel). Both human bronchial epithelial lung cells HBEC cells and THP-1-derived macrophages were exposed to different PM and analysed for cytokine release by ELISA, gene expression by qPCR and cell viability by Alamar Blue.
