      • Investigated whether a buildup of cytokines was present within the ER as a response of T cells without Sec23B in mice models, and additionally used CRISPR/Cas9 to introduce Sec23B gene deletions in human cells to study the ER response. • Utilized qPCR to determine expression levels of the unfolded protein response of mice with knockout T cells and compared to wildtype littermates. Concluded no statistical significance of upward trend in expression levels of gene expression at specified time point, however does not exclude the possibility of a difference existing at a different time point • Made use of flow cytometry and ELISA to characterize the contents in supernatant and T cells, to determine if a difference in cytokines (specifically IL2) was present in knockout and control human cells for cells with and without CRISPR/Cas9 Sec23B deletion • Dissection of mice along with culture and activation of dendritic cells and T cells, reverse transcription and qPCR activity Show less
