      BackgroundHigh-risk HPV 16 and 18 are reported to be associated in 99% of cervical cancer and many other carcinomas and HPV has reported to been present in Breast cancer. The second most prevalent type of cancer is Breast Cancer.E6 and E7 two early genes in HPV has been reported to immortalize the breast epithelial cells. Therefore this study has been carried out to investigate the association of HPV in BC.MethodsDNA is extracted from 4 paraffin embedded breast cancer tissuesusing MasterPureTMextraction kit for DNA and RNA. The extracted DNA is quantified using NanoVue technique. Highly sensitive RT-PCR was carried on to detect the presence of HPV type 16 & 56 using Amplisens HPV HCR genotype-FRT kit which is designed to test HPV DNA in clinical sample. The four fluorophores used to detect the presence were FAM (type16 &33), ROX (type18&56), HEX (type 31&35), Cy5 (β-globin).HPV type 16 and 56 is not associated with Breast Cancer but keeping in mind the positive research published, further large scale trails must be carried on to prove the association that will direct the future development of drugs. Performed routine biochemical tests involved in the project and gained expertise on various molecular biology techniques such as DNA extraction methods, Polymerase Chain Reaction (PCR), RT-PCR, RFLP, Gel electrophoresis and DNA sequencing techniques.Maintained lab notebook and other documents in accordance to the laboratory standards.Analysed test results and reported them to the project supervisor and other senior members of the team.Successfully conducted the project in accordance with agreed specifications, timelines and financial plan.
