      As part of my Bachelor's Degree curriculum, I joined Dr. John Uscian's laboratory to work on his research project: "TRYPSIN PURIFICATION, QUANTIFICATION, AND ANALYSIS OF SMOOTH TRUNKFISH (Lactophrys triqueter)". Smooth trunkfish is a species of boxfish that habits on coral reefs in the Caribbean Sea, it feeds on small mollusks, small crustaceans, sponges and tunicates. Trypsin is an enzyme that acts to degrade protein, in digestive processes it acts with the other proteinases to break down dietary protein molecules. For purification of Trypsin, we separated intestines from lumen tissueof Smooth trunkfish, followed by homogenization, centrifugation, fractured precipitation, and dialysis of the intestine sample. For quantification, we performed an enzyme assay followed by spectrophotometric procedure to determine the presence and activity of our enzyme. The assay that had most enzyme activity and presence was at the concentration of 50% of ammonium sulfate at 180s. Our results present an ideal environment in which Trypsin functions at an ideal rate. These findings can further be studied and compared in an effort to produce possible nutritional, environmental, and evolutionary connections between Smooth trunkfish at different locations across the Caribbean, or even different fish species.       As part of the ASH Minority Medical Student Award Program (MMSAP) I was able to conduct clinical research under the mentorship of Dr. Rikhia Chakraborty from the Baylor College of Medicine. I was given the opportunity to present our findings at the annual Promoting Minorities in Hematology (PMH) Event. Our goal for this project was to characterize the source of modification and immune escape strategies that caused the disease by analyzing sorted CD8/CD4 T-cell subsets isolated from lesion infiltrating cells and peripheral blood of Hodgkin Lymphoma patients. In other words, the T cells that are encountering the antigen of interest. For this purpose, we initially performed mass cytometry to subclassify the cells present in the lesion population, and then performed TCR Repertorie Sequencing to further understand the antigen mediated trigger at the deep tissue level. Once that information was gathered, we proceeded to compare it with our control group of healthy tonsil cells and analyze it using a cloud-based immune repertoire analyses system called the ClonoMap Immune Profiler. Primary limitations for mass cytometry include accessibility and availability of pediatric tissue as the main source of interest. Other limitations include that there are no specific cell lines that allows me to compare the 3 different subtypes of HL and that there are no previous proper mouse models available. On the other hand, limitations for TCR Sequencing include that there may be multiple antigens and we won’t be able to identify the exact disease-causing antigen by only using this technique since there is not a strong database.
