      Thesis title “Applications of Mesenchymal Stem Cells and Nanocarriers for Bone Tissue Engineering”Guide: Dr. Geetanjali TomarProject One - Synthesized hydroxyapatite nanoparticles (nHAp) & characterized using TEM, SEM, AFM, SAED, FTIR, DLS, & Zeta potential- Developed nanoconjugate of nHAp with PTH (1-34) & characterized for loading & release efficiency- Determined biocompatibility & differentiation potential of stem cells in the presence of the nanoconjugate- Developed osteoporotic disease model in female mice using bilateral ovariectomy & determined regenerative potential using qPCR, histology, & immunohistologyProject Two- Isolated stem cells from human gingival tissues (varying age groups)- Characterized the stem cells based on their surface markers using flow cytometry- Determined rate of proliferation, colony formation, population doubling time, senescence markers, & would healing efficiency of stem cells- Studied the effect of age on differentiation potential towards osteogenic, adipogenic, & neurogenic lineages- Evaluated the effect of age on the immunomodulatory properties of stem cells- Developed disease model of small animals (i) acute lung injury (ALI) model in mice using intranasal administration of LPS(ii) ectopic bone formation in SCID mice- Determined effect of aging on regenerative & immunomodulatory properties of stem cells in the ALI modelProject Three- Engineered thermo-responsive injectable biomaterial hydrogel & characterized it using SEM, FTIR, porosity, density, rheological analysis, & compressive strength- Designed & 3D printed PLA molds for shaping hydrogel for various biological assays- Determined biocompatibility, osteogenic, & chondrogenic potential of stem cells in the presence of hydrogel- Evaluated inflammatory response using cytokine profiling (Luminex)- Developed intervertebral disc injury model using needle puncture in rats & determined the regenerative potential of stem cell-loaded hydrogel Show less Project Title: Surface modification of UHMWPE using ECR plasma for osteoblast and osteoclast differentiation.- Ultra-high molecular Weight Polyethylene (UHMWPE) was surface modified using plasma therapy in Prof. Mathe’s Lab, Department of Physics, SPPU- Determined biocompatibility of stem cells on the surface of modified UHMWPE using MTT assay, SEM, and confocal microscopy - Evaluated differentiation potential of stem cells into osteoblast using Alizarin red S staining, Alkaline phosphatase quantification, and confocal microscopy. - Determined the effect of surface modification on the differentiation potential of peripheral blood mononuclear cells (PBMNCs) into osteoclast using confocal microscopy- Worked in collaboration with graduate students at Prof. Mathe’s Lab- Purchased equipment and consumables required for the project- Administered finances of the research grant available in the lab Show less
