PhD in Molecular and Cell BiologyThesis Title:Mechanistic Study of N-α-acetyltransferase 10 Protein in Preventing Dystocia(Manuscript in preparation)* Mastered in performing timed-pregnant, C-section, preterm birth experiments to study the "labor".* Using an ELISA, LC-MS and RT-qPCR, studied the hormonal regulation and ovarian P4 withdrawal pathways in the late gestation mice (18.5 and 19.5 d.p.c).* Using a cell-based in vitro contraction assay (3D culture), studied the uterus contraction from primary and hTERT immortalized human myometrial cells.* Using confocal, live cell imaging, In vitro acetyltransferase assays and LC-MS based methods, studied actin cytoskeleton remodeling and N-terminal acetylated substrates.Additional project:Naa10p Inhibits Beige Adipocyte-Mediated Thermogenesis through N-α-acetylation of Pgc1α.(Lee et al., and Juan, Mol. Cell. 2019, * as co-author)* Established LC-MS based In vitro acetyltransferase assay (AT assay) using recombinant purified hNaa10p.* This method has helped us to identify the key mechanistic step - N-α-acetylation status of PGC1α, PPARγ.
