  I’m characterizing the molecular function of a disordered protein under oxidative stress Acquired skills and competences:• Producing disordered proteins and research molecular mechanisms of plant stress responses in vitro and in vivo• SDS-PAGE, Western Blot, Southern Blot, electrophoresis, in-situ hybridization• Purifying protein using different chromatography techniques (ion exchange, affinity (IMAC, biotin-streptavidin, GST), size exclusion using AKTA systems.• Proteomic and protein interactome analysis to define targets related to my protein activity• Working with different membrane proteins from total cell membranes, isolated mitochondria and chloroplast to study about the interactions of my protein with its targets• Performing several assays to investigate the targets of my protein and the molecular mechanism of its activity in plant cells under stress conditions include binding kinetic such as biolayer-interference (BLI) with OctetRED 96 system and microscale thermophoresis (MST) with Nanotemper machine to derive kinetic parameter (kon, koff, kD); and enzyme kinetic In-depth analysis of enzyme activity and enzyme inhibition using Spectrophotometer, Plate reader and Stopped Flow. • Plant biotech: Agrobacterium transformation on Arabidopsis and maize, stress tests on plants, light microscope, Confocal microscopy (Zeiss, Spinning disk, Olympus), cross-breeding• Using multiple software to in study and analyze data (CLC main workbench, Graphpad prism, Pymol, Snapgene, Image J, LasX; and several sequence alignment tools, sequence analysis and structural prediction tools)• Management experience: project management; planning and performing experiment; helped organizing lab work schedule and evolving in maintenance of lab equipment.
