      Synthetic biology tools for transcriptional activation and regulation of biosynthetic gene clusters in filamentous fungi:-Modular synthetic biology toolkit for filamentous fungi-Synthetic control devices for gene regulation in Penicillium chrysogenum (rubens)-Genome editing in Penicillium chrysogenum (rubens) using Cas9 ribonucleoprotein particles-Utilization of CRISPRa/dCas9 for transcriptional regulation in filamentous fungiThe CRISPR/Cas9 system is a recent molecular tool that has revolutionized genome editing of all kinds of cells. In the CRISPR/Cas9 genome editing technology, the Cas9 endonuclease is guided by a single chimeric guide RNA to a specific locus, where it introduces a double-strand break in the DNA sequence that can be used for enhanced homology-directed repair. During the ALERT program, the aim is to employ the Cas9 protein to allow for RNA-guided transcriptional regulation. This technology will be used to upregulate and interrogate silent endogenous secondary metabolite genes in Penicillium and will be transferred to other fungi.• Collaborated on various projects related to genome editing, protein engineering, natural product identification via LC-MS, secondary-metabolite-free platform-strain engineering and anti-microbial compound discovery• Supervised and performed projects on genetic modifications, cultivations and downstream analysis• Designed experiments and genetic circuits for transcriptional regulation in filamentous fungi• Acquired skills in CRISPR/Cas9-based genome editing and transcriptional activation (CRISPRa)• Performed various downstream analysis through LC-MS, U-HPLC, prep-HPLC, qPCR, whole-genome seq.
