  During my summer research fellowship, I mastered the fundamentals of bench lab work, including pipetting skills and aseptic technique and excel data analysis. I worked closely with a mentor on the role of Tadalafil in protecting subcutaneous prostate tumors from radiation. This project required an in vitro and in vivo investigation into the mechanism of protection and allowed me to develop a proficiency in various in vitro cell culture techniques. I isolated DNA, RNA, and protein to look at PDE5 (the target of Tadalafil) expression as well as performed multiple clonogenic survival assays of two prostate cancer cell lines (DU 145 and PC-3) exposed to varying concentrations of Tadalafil one hour prior to radiation therapy. Concurrently, I studied the effects of Tadalafil on subcutaneous prostate cancer growth. Mice were treated for five days with Tadalafil one hour prior to radiation and tumors were measured multiple times a week for the course of the study. I assisted in collecting subcutaneous and orthotopic tumors at the end of various in vivo studies. Show less
