The goal was to in silico study the steady state expression pattern of the genes involved in the circuit within a time frame.       Project title: STAT3 A Key Regulator and Novel Therapeutic Target in Osteopontin – Induced Tumor Growth and Angiogenesis in Breast CancerLab: Signal Transduction Lab, NCCS, PuneSupervisor: Dr. Gopal Kundu, Scientist G – Shanti Swaroop Bhatnagar AwardeeResearch Description: The project planned to investigate the angiogenic role of osteopontin in breast cancer progression. My results showed an increase in the tumour volume in NOD/SCID mice injected with osteopontin induced MCF – 7 cell line in comparison with the parental MCF – 7 cell line. Immunohistochemistry results of the respective angiogenesis specific molecules correlated with animal experiment results. We also performed cell proliferation assays to examine combinatorial effects of novel STAT3 inhibitor and tamoxifen on osteopontin induced breast tumour cell proliferation. Show less

      Project Title: Profiling the Incidence of Novel Alterations Discovered in Human Lung CancerLab: Integrated Cancer Genomics Lab, ACTREC – a research wing of Tata Memorial Hospital (TMH), Mumbai, IndiaSupervisor: Dr. Amit Dutt, Scientist F (Assistant Professor/Wellcome – DBT India Alliance Trust Intermediate Fellow)Research Description: Integrated genomic analysis performed by my peers upon integrating the data obtained from four head and neck cancer cell lines pointed out NRBP1 as novel candidate. My work focused on evaluating the oncogenic potential of this novel heterozygous truncating pseudokinase domain mutation (Q73*) within Nuclear receptor binding protein (NRBP1) gene using cellular and molecular approaches. My results strongly indicated that NT8e cell line harboring mutant NRBP1 gene is oncogenic as it drives the immortalized cells (NIH3T3) to form soft agar colonies. shRNA knockdown of mutant NRBP1 in NT8e cell line significantly inhibited anchorage – independent growth and demonstrated its essentiality for oncogenic transformation.
