Live functional anatomy was controlled under… Show more Published Thesis: Ex-Vivo Slaughterhouse Porcine Crystalloid-Perfused Beating Heart via Langendorff Method:The objective of this study was to resuscitate isolated beating porcine hearts obtained from slaughterhouse swine for a minimum of 1 hour with a crystalloid buffer reperfusion instead of blood via an extracorporeal Langendorff apparatus. The isolated beating heart functions outside of the body under simulated physiologic conditions. Live functional anatomy was controlled under benchtop experimental settings. Porcine hearts (26 total) were preserved and reanimated with an oxygenated modified Krebs Henseleit buffer perfusion. Low warm ischemic time, Streptokinase, and coronary catheterization resulted in 1 hour global contraction and mean 110bpm stabilized sinus rhythm. ECG displayed prominent QRS complex.       From April 2013, I initiated the design for the Langendorff model based on several literatures under the guide of Dr. Azadani for my Masters Thesis. The current goal is an experimental validation study in which fresh preserved porcine heart is to be resuscitated towards near-physiological conditions after 1-2 hour ischemic time. The research encompasses varied tasks of different disciplines such as the following: looking through literature such as the buffer preparation, networking and seeking advice from authors of the literature as well as cardiac surgeons and heart transplant administrators, field work of collecting the porcine heart, and drug delivery and pacing while testing. Research includes comparisons in both Langendorff vs. working heart model as well as cardioprotective buffers, ordering equipment and pharmaceuticals, system design, organ/heart preservation, water quality, modified buffer preparation to exactly pH 7.4.Future goals include testing heart valves on the beating resuscitated porcine heart, testing medical devices as well as testing on human heart both from University of Colorado Denver Anschutz Medical Campus.
