Then, using the state-of-the-art TrAEL-seq method, we found evidence of a profound disruption of the replication in chronologically aged S. cerevisiae; during my PhD, I will use techniques such as the circle-seq, OG-seq and polymerase-usage sequencing to investigate the causes of this disruption and its importance for the process of ageing.
