      At this position I was working in the research team on the ongoing projects in laboratory.• Characterization and further development of a new optical approach to measure fast redox signals with single-organelle resolution in living mice that express genetically-encoded redox biosensors in their neuronal mitochondria (Breckwoldt et al., Nature Medicine 2014).• To study microtubules in intact mammalian neurites, we devised a new imaging assay based on transgenic mice expressing yellow fluorescent protein (YFP) tagged end-binding protein 3 (EB3). This approach allows measuring microtubule dynamics in vivo and in mature neurites in the PNS and CNS under physiological conditions and after exposure to microtubule-modifying drugs (Kleele et al., Nature Communications 2014). Show less In my PhD research I used in vivo microscopy to better understand axonal transport deficits which are widely assumed to be key pathogenic event in ALS and other neurodegenerative diseases. Contrary to expectations, I discovered a dissociation of axonal degeneration and transport deficits in SOD-based ALS models (Marinkovic et al., PNAS 2012). 1. Used wide-filed, confocal and two-photon microscopy to follow transport of fluorescently labelled mitochondria and endosomes in mouse models of ALS.2. Developed experimental technique to follow transport of individual mitochondria using novel mouse transgenic line Thy1-MitoKaede. This approach allows studying of transport characteristics and following destiny of single mitochondria over long distances in axons.3. Manuscript preparation, submission and revision to the peer-reviewed journal and presentation of research at seminars and international conferences. Show less

      Alzheimer's disease is characterised by cognitive decline driven by malfunctioning and degeneration of neurons. To better understand these changes, I developed an approach which combines two-photon calcium imaging through cranial window with imaging in mouse models of Alzheimer’s disease. Using this technique I am now able to follow same neuronal populations over prolonged periods of time and identify when during the disease course first impairments in neuronal activity occur.
